Dipeptide Diaminobutyroyl Benzylamide Diacetate (commonly known as SYN-AKE or Snake Peptide) is a synthetic peptide used in high-performance anti-aging cosmetics. It is a biomimetic tripeptide designed to mimic the activity of Waglerin-1, a neurotoxic peptide found in the venom of the Temple Viper (Tropidolaemus wagleri). Despite its origin inspiration, the synthetic version is safe, non-toxic, and provides a topical alternative to injectable neurotoxins like Botox.
Unlike Acetyl Octapeptide-3 (which targets the SNARE complex pre-synaptically), Dipeptide Diaminobutyroyl acts on the post-synaptic membrane.
Receptor Binding: It acts as a reversible antagonist of the muscular nicotinic acetylcholine receptor (nmAChR).
Blocking the Signal: Specifically, it binds to the ε-subunit of the receptor, preventing acetylcholine (the neurotransmitter that signals muscles to contract) from binding.
Muscle Relaxation: With the receptor blocked, the ion channel remains closed. Sodium ions (Na⁺) cannot enter the cell, preventing depolarization. The nerve excitation transmission is blocked, and the facial muscles remain relaxed.
