  • Description

Dipeptide Diaminobutyroyl Benzylamide Diacetate (commonly known as SYN-AKE or Snake Peptide) is a synthetic peptide used in high-performance anti-aging cosmetics. It is a biomimetic tripeptide designed to mimic the activity of Waglerin-1, a neurotoxic peptide found in the venom of the Temple Viper (Tropidolaemus wagleri). Despite its origin inspiration, the synthetic version is safe, non-toxic, and provides a topical alternative to injectable neurotoxins like Botox.

Mechanism of Action: How It Works

Unlike Acetyl Octapeptide-3 (which targets the SNARE complex pre-synaptically), Dipeptide Diaminobutyroyl acts on the post-synaptic membrane.

  1. Receptor Binding: It acts as a reversible antagonist of the muscular nicotinic acetylcholine receptor (nmAChR).

  2. Blocking the Signal: Specifically, it binds to the ε-subunit of the receptor, preventing acetylcholine (the neurotransmitter that signals muscles to contract) from binding.

  3. Muscle Relaxation: With the receptor blocked, the ion channel remains closed. Sodium ions (Na⁺) cannot enter the cell, preventing depolarization. The nerve excitation transmission is blocked, and the facial muscles remain relaxed.

Result: The intensity of muscle contractions is reduced, leading to a visible softening of dynamic wrinkles (expression lines) such as crow's feet, forehead lines, and laugh lines.

Efficacy & Clinical Data

Clinical studies have demonstrated the potency of this peptide:

  • Wrinkle Reduction: A study involving 15 volunteers (aged 40-60) using a 4% SYN-AKE cream twice daily for 28 days showed a 52% reduction in wrinkles.

  • Consumer Feedback: In the same study, 80% of participants reported a smoothing effect, and 73% reported visibly reduced wrinkles, with particularly good results for deeper wrinkles in older adults.

  • Potency: It is often cited as being 5 times more effective at reducing dynamic wrinkles than Argireline (Acetyl Hexapeptide-8), though efficacy varies by formulation.

Formulation Guidelines

Parameter Recommendation
Recommended Usage 0.5% – 5.0% (typical cosmetic formulations); up to 4% (maximum historical use in leave-on products)
pH Stability Stable at pH 4.0 – 7.0
Incorporation Temperature Below 40°C (104°F) during the final stage of manufacturing
Storage -20°C (Long-term); 2-8°C (Short-term). Keep tightly closed and dry

Applications in Skincare

Due to its potent "Botox-like" effect without needles, it is widely used in:

  • Anti-aging serums and essences

  • Eye creams (targeting crow's feet)

  • Facial masks and patches

  • Neck and décolleté treatments

  • Day and night anti-wrinkle creams
